Operation and calibration of UV cabinet

PURPOSE : To provide a procedure for operation and calibration of UV Cabinet.

SCOPE : This SOP is applicable for quality control department for operation and calibration of UV Cabinet.

RESPONSIBILITY

Analyst / QC – officer / Executive – QC : To operate and calibrate the instrument as per SOP.

Manager – QC :  To ensure implementation of SOP.

Head QC : To ensure implementation of SOP.

PROCEDURE

Cleaning

Disconnect the instrument from main power supply.

 Clean the apparatus accessories with the help of clean dry lint free cloth or suitable brush daily.

 Operation

Plug the cord of the UV-cabinet in plug and Switch on the main electrical switch.

There are three liver located on the instrument having written near them On UV-Long (365nm), On UV-Short (254nm) and On Visible. When you pull respective liver forward the respective UV and Visible light glows.

Now pull the cabinet door and put the TLC plate at right angle to the direction of light, shut the door and on the respective source which mention in the monograph and observe the plate through the window given on the top of cabinet.

When operation over switch of the source light and then switch off the main switch and take out TLC Plate. Record the activity in log book.

Calibration

Material required

  • Sodium Salicylate
  • Ethanol (95%)
  • Silica gel-G
  • TLC Plate
  • Purified Water.

Silica gel-G Plate

Readymade silica gel-G plate used.

Preparation of ethanol (95%)

Take about 5ml of water in 100ml volumetric flask and make up to volume with ethanol (99.99%).

Preparation of 2% w/v Sodium Salicylate Solution (Solution-1)

Weight about 200mg of Sodium Salicylate and transfer it into 100ml volumetric flask add sufficient ethanol (95%) to dissolve and make up to volume with same solvent.

 Preparation of 04% w/v solution of sodium salicylate

Pipette 10ml from solution-1 and transfer to 50ml volumetric flask and make up to volume with ethanol (95%)

 Method

Operate the apparatus according to procedure

Apply to a plate coated with silica gel G, 5µl of a 0.04% w/v solution of sodium salicylate in ethanol (95%) for lamps of maximum output at about 254nm and 5µl of a 0.2% w/v solution of sodium salicylate in ethanol (95%) for lamps of maximum output at about 365nm. Examine the spot in a position normal to the radiation. The distance between the lamp and the plate under examination used in a pharmacopoeia test should not exceed the distance used to carry out the above test.

 Record the Observation

If parameter does not meet the specified limits then immediately paste the label “Out of Calibration, Do not Use” on the Instrument and Inform QC Head.

Acceptance criteria

The lamp should be capable of revealing Without doubt a standard spot of sodium salicylate with a diameter of about 5mm on a chromatographic plate coated with silica gel G.

Frequency : 3 Months

Precautions

Make sure the TLC Plate is properly place in the UV cabinet.

Ensure the cleanliness of the instrument.
